ABSTRACT


Dairy products such as yogurt and locally made cheese are nutrient-rich foods prone to microbial contamination, which affects quality, safety, and shelf life. Microorganisms including bacteria, yeasts, and molds are responsible for spoilage and may pose health risks. The purpose of this study is to isolate and identify microorganisms responsible for the spoilage of yogurt and locally made cheese, and to assess their abundance and diversity. This research was motivated by frequent reports of spoiled dairy products in Nigeria, which lead to economic loss and potential health hazards.

Samples of yogurt and locally made cheese were collected from Lagos markets. Microbial isolation was performed using selective media, followed by morphological, biochemical, and microscopic identification. Colony counts were determined to quantify microbial load. The findings show yogurt had bacterial counts of 4.9 × 105– 5.6 × 105CFU/mL dominated by Lactobacillus bulgaricus and Streptococcus thermophilus, while cheese had 3.8 × 106– 4.2 × 106CFU/g with Lactobacillus plantarum and molds like Aspergillus sp. Furthermore, spoilage organisms such as Staphylococcus aureus and Candida sp. were detected.

The outcome of this research indicates that both yogurt and locally made cheese harbor beneficial and spoilage microorganisms, emphasizing the need for proper hygiene, controlled fermentation, and safe storage to maintain quality and protect consumer health.Based on the findings of this study, it was recommended that dairy producers and handlers should maintain strict hygienic practices during the production of yogurt and locally made cheese to minimize contamination by pathogenic and spoilage microorganisms. Also, raw materials, especially milk, should be properly pasteurized before fermentation to reduce the risk of bacterial and fungal contamination.



1.1 Introduction

Microorganisms are microscopic organisms, including bacteria, fungi, and yeasts, that are present in virtually all environments and play significant roles in both beneficial and detrimental processes (Madigan et al., 2021). In the context of food, microorganisms are essential for fermentation but are also major contributors to spoilage when their growth is uncontrolled. Dairy products, particularly yogurt and locally made cheese, are highly nutritious foods widely consumed for their rich protein content, calcium, and probiotic benefits (Adetunji et al., 2020).

Spoilage of dairy products is a significant concern in the food industry because it leads to economic losses, reduced shelf life, and potential health risks for consumers. The microbial contamination of yogurt and locally made cheese is influenced by several factors, including poor handling practices, inadequate sanitation during production, and improper storage conditions (Ibrahim et al., 2021). Common spoilage microorganisms include lactic acid bacteria, molds, yeasts, and pathogenic bacteria such as Escherichia coli, Staphylococcus aureus, and Salmonella species, which can alter the taste, texture, and safety of the products (Olajuyigbe & Afolabi, 2019).

As a prelude to other parts of this study, this chapter will discuss the background upon which this study was initiated, the statement of problems that led to this study, the Aim and Objectives of the study. Others are significance of the study, scope of work, limitation of the study and definition of terms.


1.2 Background of Study

The study of microorganisms in food dates back to the late 19th century when pioneers such as Louis Pasteur established the relationship between microbes and food spoilage. Pasteur affirmed that microbial activity is responsible for fermentation and deterioration in perishable products, including dairy items. The understanding of microbial contamination has since evolved, leading to the development of food microbiology as a distinct scientific discipline (Madigan et al., 2021). Yogurt and cheese have long histories as staple dairy products, with fermentation playing a central role in their production. Fermented dairy products were traditionally made using natural starter cultures, which contain a mixture of lactic acid bacteria that preserve the product and impart desirable flavors (Olajuyigbe & Afolabi, 2019).

In Nigeria, locally made cheese and yogurt have been produced for decades, relying primarily on artisanal methods that often lack standardized quality control measures. Researchers have asserted that these products are particularly susceptible to contamination by spoilage microorganisms, including Lactobacillus, Staphylococcus, and various yeasts and molds, which thrive in high-moisture and nutrient-rich environments (Ibrahim et al., 2021). It has also been reported that inadequate refrigeration, poor handling practices, and exposure to environmental contaminants exacerbate the rate of spoilage in these products.

Dairy products, including yogurt and locally made cheese, are important sources of nutrition worldwide due to their high content of proteins, calcium, vitamins, and beneficial microorganisms such as probiotics. According to Adetunji et al. (2020), the consumption of these products contributes significantly to human health, particularly in improving digestive function and strengthening the immune system. However, these products are highly perishable and susceptible to microbial spoilage because of their rich nutrient composition and high moisture content.

It has been reported that microbial contamination is a major factor leading to the deterioration of dairy products, resulting in changes in taste, texture, color, and overall quality (Ibrahim et al., 2021). Studies have also asserted that both beneficial and pathogenic microorganisms can influence the shelf life of dairy products, with uncontrolled microbial growth posing significant health risks to consumers. For instance, Olajuyigbe and Afolabi (2019) stated that bacteria such as Escherichia coli, Staphylococcus aureus, and molds like Aspergillus species are frequently responsible for spoilage in dairy products, leading to foodborne illnesses if consumed.

Researchers have affirmed that inadequate hygiene practices during production, processing, and storage contribute heavily to microbial contamination (Okonko et al., 2018). It is further contended that locally made cheese is particularly vulnerable due to its traditional processing methods, lack of standardized procedures, and minimal cold-chain storage, which favor the growth of spoilage microorganisms. According to Madigan et al. (2021), systematic isolation and identification of these microorganisms are critical for understanding the factors that affect spoilage, implementing preventive measures, and improving the safety and quality of dairy products. This study is set against the backdrop of the need to understand the specific microorganisms responsible for the spoilage of dairy products, the conditions that promote their growth, and the strategies that can be employed to mitigate contamination in both industrial and locally produced dairy items.


1.3 Statement of Problems

Investigation revealed that dairy products such as yogurt and locally made cheese are widely consumed due to their nutritional benefits, including high protein content, essential vitamins, and probiotics that promote gut health (Adetunji et al., 2020). However, these products are highly perishable and susceptible to microbial contamination (Olajuyigbe & Afolabi, 2019).

Additionally, the inability of producers to accurately identify the microorganisms responsible for spoilage is a significant problem. Without proper identification, it is difficult to develop effective preservation methods or ensure product safety. On the other hand, inadequate hygienic practices during production, processing, and storage further exacerbate microbial contamination, making dairy products unsafe for consumption and limiting their shelf life (Ibrahim et al., 2021).

Furthermore, spoilage not only affects commercial profitability but also has public health implications. Contaminated dairy products may harbor pathogenic microorganisms that cause foodborne illnesses, particularly among vulnerable populations such as children, the elderly, and immunocompromised individuals (Okonko et al., 2018). It is against this backdrop that this study seeks to isolate and identify the microorganisms responsible for the spoilage of yogurt and locally made cheese.


1.4 Aim and Objectives of Study

The aim of this study is to isolate and identify the microorganisms responsible for the spoilage of yogurt and locally made cheese, and to assess their impact on product quality and safety. To achieve this aim, the study has the following objectives:

  1. To determine the types of microorganisms present in spoiled yogurt and locally made cheese.
  2. To identify the specific bacteria, yeasts, and molds responsible for spoilage.
  3. To assess the factors that favor microbial growth in these dairy products.
  4. To recommend measures for reducing spoilage and enhancing the safety and shelf life of dairy products.

1.5 Research Questions

The study came up with research questions so as to be able to ascertain the above stated objectives. The specific research questions for the study are stated below as follows:

  • What types of microorganisms are present in spoiled yogurt and locally made cheese?
  • Which specific microorganisms are responsible for the spoilage of these dairy products?
  • What factors contribute to the growth of spoilage microorganisms in yogurt and locally made cheese?
  • What measures can be implemented to reduce spoilage and improve product safety and shelf life?

1.6 Significance of Study

It is believed that at the completion of the study, the findings will assist dairy producers, both industrial and artisanal, in implementing effective hygiene practices and storage techniques to reduce spoilage and extend the shelf life of their products. This study will also be beneficial to regulatory bodies and food safety authorities by providing scientific data that can guide the development of standards and policies aimed at ensuring the safety and quality of dairy products.

Additionally, the research will support the economic sustainability of dairy businesses by minimizing losses due to spoiled products, enhancing consumer confidence, and promoting better market competitiveness.

Furthermore, it will contribute to public health by reducing the risk of foodborne illnesses associated with contaminated dairy items, thereby protecting consumers, particularly vulnerable populations such as children, the elderly, and immuno-compromised individuals.

Lastly, the study will serve as a reference point for future studies on microbial spoilage in dairy products, fostering further research and innovation in food microbiology and preservation techniques.


1.8 Scope of Study

The scope of the research is focused on the isolation and identification of microorganisms responsible for the spoilage of yogurt and locally made cheese in Lagos State, Nigeria. Specifically, the research targets products obtained from selected commercial producers and local markets, including Eko Foods Limited in Lagos, to represent both industrial and artisanal production systems. The study will examine microbial types, growth factors, and contamination sources.


1.9 Limitations of the Study

During the course of this study, there were some problems encountered which stood as limitations to the research work. Some of the limitations include:

  1. Time Constraint: The time frame given to accomplish this project was very short due to school academic calendar and it was carried out under pressure which made the researcher not to implement some necessary features.
  2. Financial Constraint: Insufficient fund tends to impede the efficiency of the researcher in sourcing for the relevant materials, literature or information and in the process of data collection (internet, questionnaire and interview).
  3. Initial Cooperation Delay from Respondents: A particular limitation of this work came as a result of the respondent refusal to offer their cooperation at the initial time they were contacted. This contributed in making the success of this research study difficult.

1.10 Definition of Terms

Dairy Products: Foods derived from milk, including yogurt and cheese, that are rich in proteins, calcium, and other nutrients (Adetunji et al., 2020).

Yogurt: A fermented dairy product produced by the bacterial fermentation of milk, primarily using Lactobacillus and Streptococcus species (Olajuyigbe & Afolabi, 2019).

Locally Made Cheese: Cheese produced using traditional or artisanal methods within local communities, often without standardized procedures or industrial equipment (Ibrahim et al., 2021).

Microorganisms: Microscopic organisms such as bacteria, yeasts, and molds that are present in the environment and food products, some of which contribute to spoilage or fermentation (Madigan et al., 2021).

Spoilage: The process by which food deteriorates due to microbial activity, resulting in changes in taste, odor, texture, and safety (Okonko et al., 2018).

Isolation: The process of separating specific microorganisms from a mixture for identification and study (Madigan et al., 2021).
